4Cell® SmartCHOpe media 는 N-1 perfusion 공정에 효율적으로 사용됩니다 (Figure 1A).
CHO cell 은 100 million cells/mL 의 고밀도 perfusion cell culture 를 5-7 일동안 진행할 수 있습니다 (Figure 1B).
그 후 200리터 bioreactor 에서 대용량 세포배양을 할 수 있는데 이는 4Cell® SmartCHOpe media 는 standard seed train 에서 탁월한 세포성장과 viability 를 보여주고 있다는 것을 의미합니다 (Figure 1C).

4Cell® SmartCHOpe media 는 연속적인 perfusion applications 을 할 수 있습니다. 오른 쪽 data 는 ATF device 와 연결된 2 Liter Bioreactor 에서 4Cell® SmartCHOpe media 를 이용하여 continuous perfusion 방식으로 세포를 키운 결과를 보여주고 있습니다. 세포는 고밀도, 높은 세포생존율과 생산성을 유지하면서 34일동안 세포배양을 진행할 수 있었다.
Note. Viable cell density (VCD) and percentage viability were measured in a 2 L bioreactor with ATF for 34 days.

N-1 perfusion 에 의한 high innoculation fed-batch 를 이용하면 CHO clone 에 따라 10 g/L 까지 생산성을 증가시킬 수 있습니다.
Note. N-1 perfusion: cultivation was performed in the Biostat® RM System with 4Cell® SmartCHOpe at 1 L working volume. High-inoculation fed-batch cultivation was carried out in 5 L UniVessel® with 4Cell® SmartCHOpe as basal medium, and 1.3× 4Cell® SmartCHO feeds added to provide adequate nutrient supply throughout the whole process
