Purpose

ÀÌ application note ´Â Sartorius BIOSTAT¢ç 2L single-use Univessel ¹è¾ç±â¿¡¼­ human adipose-derived mesenchymal stem cells (hAD-MSC) ¼¼Æ÷¸¦ microcarrier ¿¡¼­ cell expansion ÇÒ ¶§ÀÇ ¹è¾çÁ¶°Ç°ú ¼³Á¤°ª¿¡ ¼³¸íÇÏ°íÀÚ ÇÑ´Ù.

ÀϹÝÀûÀ¸·Î ÀÌ·¯ÇÑ ¹è¾çÁ¶°ÇÀº 50 Liter ÀÌ»ó±îÁö scale up ÇÏ¿© ¹è¾çÇÒ ¼ö ÀÖ´Ù. ½Å·Ú¼ºÀÌ ÀÖÀ¸¸ç ¶ÇÇÑ scale up µµ °¡´ÉÇÑ cell expansion ¹æ¹ýÀº MSC ¼¼Æ÷¸¦ Ä¡·áÁ¦·Î »ç¿ëÇϴµ¥ ¸Å¿ì Áß¿äÇÑ ³»¿ëÀÌ´Ù.


Introduction

Medium, Methods & Equipment

Overview of set-up procedure

Day -1 : microcarrier bead ÀÎ Sartorius SoloHill Pronectin F ¸¦ Àüü ¹è¾ç¾×ÀÇ 0.75% (solid fraction) ³óµµ·Î ¹Ì¸® ÁغñÇÑ´Ù.
 
Day 0 : ÁغñÇÑ UniVessel SU bioreactor ¿¡ microcarrier ¸¦ ³Ö°í MSC ¹è¾ç¾×À» 0.7 Liter ä¿î´Ù. ±×¸®°í 4½Ã°£µ¿¾È bioreactor ¿¡ ÀåÂøµÈ pH & DO ¼¾¼­¸¦ equilibration ÇÑ´Ù.

³Ãµ¿µÇ¾î ÀÖ´Â MSC ¼¼Æ÷¸¦ ³ìÀÎ ÈÄ seeding density °¡ 1.5 x 10^7 °¡ µÇµµ·Ï ÁغñÇÑ ÈÄ bioreactor ¿¡ ³Ö°í ÀÌ ¼¼Æ÷µéÀÌ microcarrier ¿¡ ºÙÀ» ¼ö ÀÖµµ·Ï agitation ¾øÀÌ 4½Ã°£ µ¿¾È ±×´ë·Î µÐ´Ù.

±× ÈÄ bioreactor ÀÇ media ¾çÀ» ÃÑ 2 Liter ·Î ¸ÂÃß°í 100rpm ÀÇ ¼Óµµ·Î agitation À» ½ÃÀÛÇÑ´Ù.
 
Day 4 : bioreactor ¾ÈÀÇ ¹è¾ç¾× 50% ¸¦ fresh media ·Î ±³Ã¼ÇÑ´Ù.
 
Day 7 : enzymatic cell detachment => cell resuspension => vialing => freezing ÀÇ ¼ø¼­·Î microcarrier ¿¡ ºÙ¾îÀÖ´Â MSC ¼¼Æ÷¸¦ ºÐ¸®ÇÑ´Ù.




Cells, Medium and Microcarrier

Ãʱâ seeding Çϴµ¥ »ç¿ëÇÑ 1.5 x 10^7 cell Àº serum-reduced 5% FBS media ¿¡¼­ ¹è¾çÇÏ¿´´Ù. SoloHill Pronectin F microcarrier (0.75% solid fraction) Àº Á¦Á¶»ç ±ÇÀå ¹æ¹ýÀ¸·Î ÁغñÇÏ¿´´Ù.

Cultivation setup

´ÙÀ½Àº UniVessel Su ¿¡¼­ MSC ¼¼Æ÷¸¦ cultivation ÇÏ´Â 3°¡Áö °úÁ¤ÀÌ´Ù. (1) initial cell attachment (2) cell expansion (3) cell harvest

Microcarrier ¿Í 0.7Liter ¹è¾ç¾×À» UniVessel SU bioreactor ¿¡ ³Ö°í 37¡É, 5% CO©ü, 85 rpm ÀÇ Á¶°Ç¿¡¼­ 4½Ã°£µ¿¾È equilibration ÇÑ´Ù.

±× ÈÄ Çص¿ÇÑ MSC ¼¼Æ÷¸¦ Ãʱâ density °¡ 2.8 x 10^3 cells/cm©÷ °¡ µÇµµ·Ï ³Ö¾îÁÖ°í 4½Ã°£µ¿¾È agitation ¾øÀÌ ±×´ë·Î ¹æÄ¡ÇÏ¿© ¼¼Æ÷°¡ microcarrier ¿¡ attachment Çϵµ·Ï ÇÑ´Ù.

UniVessel SU vessel ÀÇ ¹è¾ç¾×ÀÌ ÃÑ 2 Liter °¡ µÇµµ·Ï ¹è¾ç¾×À» ä¿öÁØ´Ù.

Ãʱ⿡´Â impeller ÀÇ speed ¸¦ 100rpm À¸·Î ½ÃÀÛÇÏ°í ¹è¾çÀÌ ÁøÇàµÊ¿¡ µû¶ó ÃÖ´ë 135rpm À¸·Î ¿Ã·ÁÁØ´Ù.

MSC ¼¼Æ÷´Â 37¡É, pH 7.2 ±×¸®°í 0.1 vvm (gas volume flow per unit of liquid volume per minute) Á¶°Ç¿¡¼­ 7ÀÏ µ¿¾È ¹è¾çÇÏ¿´´Ù.

¹è¾ç 4ÀÏ°¿¡ (cell density °¡ ¾à 2 - 3 x 10^4 cells/cm©÷) ¿µ¾çºÎÁ·À» ¹æÁöÇϱâ À§ÇØ vessel ³» ¹è¾ç¾× 50% ¸¦ »õ ¹è¾ç¾×À¸·Î ±³Ã¼ÇÏ¿© ÁØ´Ù.

Cultivation conditions

- working volume : 2 L
- Microcarrier concentration : 0.75% microcarrier soilid fraction
- Inoculated cell number : 1.5 x 10^7 MSC
- Agitation speed : between 100rpm (below NS1u) and 135rpm(NS1)
- Temperature : 37¡É
- Head aeration : 0.1 vvm
- Dissolved oxygen (DO) : > 20%
- Cultivation time : 7 days


Results

¹è¾ç 4ÀÏ°¿¡ ¹è¾ç¾×Áß 50% »õ°ÍÀ¸·Î ±³È¯ÇÏ¿´´Ù.

¹è¾ç 7ÀÏ°¿¡ ¼¼Æ÷¸¦ harvest Çϱâ Á÷Àü¿¡ DAPI staining À» ½Ã½ÃÇÏ¿© °üÂûÇÑ °á°ú ¸ðµç microcarrier °¡ ¼¼Æ÷¿¡ ¿ÏÀüÈ÷ µÑ·¯½×¿© ÀÖ¾úÀ¸¸ç ±× aggregates ÀÇ Á÷°æÀº ¾à 2mm Á¤µµ¿´´Ù.

Trypsin EDTA ¸¦ ÀÌ¿ëÇÏ¿© ¼¼Æ÷¿Í microcarrier ¸¦ ºÐ¸®ÇÑ ÈÄ ¼¼Æ÷¸¦ harvest ÇÏ¿´À¸¸ç harvest rate ´Â > 98%, cell viability ´Â 98% À̾ú´Ù.


Conclusion
