Introduction

¿ìÀ¯¿Í °°Àº À¯Á¦Ç°µéÀº ÀÎü¿¡¼­ ¸Å¿ì Áß¿äÇÑ ±â´ÉÀ» ¼öÇàÇÏ°í ¶Ç ±ÕÇüÀâÈù ¿µ¾ç¼·Ãë¿¡ ÀÖ¾î ÇʼöÀûÀÎ ±â´ÉÀ» ¼öÇàÇÑ´Ù. Áö¹æ, mineral, lactose ±×¸®°í ´Ü¹éÁú°ú ¿ìÀ¯ ¼ººÐÀº ¿ìÀ¯ ¼ººÐºÐ¼®¿¡¼­ ¸Å¿ì Áß¿äÇÑ ¼ººÐµéÀÌ´Ù.

Protein Determination in milk according to the Kjeldahl method

ų´Þ (Kjeldahl) Àº ½ÄÇ°°ú »ç·áÀÇ Áú¼Ò¿Í ´Ü¹éÁú ÇÔ·® ºÐ¼®ÀÌ ¸Å¿ì Á¤È®ÇÏ°í ÀçÇö¼ºÀÌ ¸Å¿ì Ź¿ùÇÏ¿© ¿À´Ã³¯ ³Î¸® »ç¿ëµÇ°í ÀÖ´Ù.
Çö´ëÀÇ Å³´Þ¹ýÀº Á¦ÀÏ ¸ÕÀú À¯±â¹°ÁúÀ» Ȳ»ê¿¡ ³Ö°í 400¡ÆC ÀÌ»óÀÇ °í¿Â¿¡¼­ °¡¿­Çϴµ¥ ÀÌ °á°ú »ùÇüӿ¡ À¯±âÀûÀ¸·Î °áÇյǾî ÀÖ´ø Áú¼Ò°¡ ammonium sulfate ·Î º¯È¯µÇ°Ô µÈ´Ù
ÀÌ ¾Ï¸ð´½¿°À» alkali ¿ë¾×À¸·Î Áßȭó¸®ÇÏ¿© NH3 ·Î ¸¸µé°í À̸¦ steam À¸·Î distillation ÇÏ¿© ¾Ï¸ð´Ï¾ÆÀÇ ¾çÀ» Á¤·® (titration) ÇÏ°Ô µÈ´Ù.

Sample

- Liquid bovine semi-skimmed milk
- Protein labeled value: 3.25 g/100 ml

Sample Digestion

- Stir the milk into a beaker using a VELP magnetic stirrer for 60 sec. at 700 rpm.
- Put 5 ml of sample into a 250 ml test tube, by using a pipette.
- For each sample, add in the test tube:
  * 2 catalyst tablet CM (code CT0006650; 3.5 g K2SO4, 0.1 g CuSO4, 5H20 Missouri)
  * 20 ml concentrated sulphuric acid (96-98%)
  * 5 ml of hydrogen peroxide (~ 30%)
- Prepare some blanks with all chemicals and without sample.
- Connect the Digestion Unit to a proper Aspiration Pump (JP code F30620198)
  and a Fume Neutralization System (SMS Scrubber code F307C0199)
  to neutralize the acid fumes created during digestion phase.
- Digest the samples for 15 minutes at 150 ¡É, plus 15 minutes at 250 ¡É and
  40 minutes at 420 ¡É according to the method ¡°milk and derived products¡± (n¡Æ 1 on DKL 20).

Distillation and Titration

- Let the test tubes cool down to 50-60 ¡É.
- Condition the UDK 159 unit by performing the Automatic Check up in Menu-System
  and a Wash down.
- Distill the samples according to the following parameters (pre-defined method n¡Æ1):
  * H2O (dilution water): 50 ml
  * H2SO4 (0.1 N) as titrant solution
  * NaOH (32 %): 70 ml
  * Protein factor: 6.38
  * H3BO3 (4 % with indicators): 30 ml
- Distillation & Titration analysis time: from 4 minutes for one test.

Typical Results on Semi-Skimmed Milk

- The results are automatically calculated by UDK 159 as a percentage of nitrogen
  and percentage of proteins.
- This is ¡°protein¡± on a total nitrogen basis.
